Temperature and substrate dependence of peroxidase activity in the extract from omorika needles (Picea omorika (Panc.) Purkinye)

We studied temperature dependence of POD activity in the extract from omorika needles (Picea omorika (Panc.) Purkinye). Chosen temperatures were in physiological range. We used two substrates - guaiacol and coniferyl alcohol, and measured POD activity using UV - VIS spectrophotometry. In case of both substrates there was an increase in maximal velocity of the enzyme reaction with temperature at lower temperature range. At the temperature 30 deg C POD maximal velocity in reaction with guaiacol attained a maximum, while activity with coniferyl alcohol showed a permanent increase.
