Analysis of 12 microsatellite loci in Bali cattle genome

Polymorphisms of 12 microsatellite markers (BM 2113, CSSM 66, ETH 10, ETH 152, ETH 152, HEL 1, HEL 5, ILSTS, INRA 023, INRA 032 and INRA 035) have been studied in Bali cattle (Bos sondaicus). DNA samples extracted from 25 Bali cattles were used as prominent materials PCR amplifications have been done using 12 pairs of primer flanking the above microsatellites. PCR products were separated by 6 percent polyacrylamide gel electrophoresis. The silver staining method was used to detect allels of each locus. The result showed that from 12 microsatellites loci analyzed, the average number of allels was 1.83, with the highest number of allel was 3 and the lowest number of allel was 1. Low genotype variability within Bali cattle population suggested that Balk breed has level of purity

Amplifikasi PCR dilakukan menggunakan 12 pasang primer pengapir mikrosatelit. Produk PCR dipisahkan dengan elektroforesis gel poliakrilamid 6 persen. Metode pewarnaan perak digunakan untuk mendeteksi alel setiap lokus. Hasil penelitian menunjukkan bahwa dari 12 lokus mikrosatelit yang dianalisis, rata-rata alel yang diperoleh 1,83 buah, dengan jumlah alel tertinggi 3 dan terendah 1. Variasi genotipe yang rendah pada populasi sapi Bali menunjukkan bahwa bangsa sapi Bali masih memiliki kemurnian tinggi
