Studies on retrieval rate and biometrics of oocytes from ovaries of slaughtered murrah buffaloes

The follicular contents of 76 ovaries from 38-slaughtered Murrah buffaloes were aspirated and dispensed in phosphate buffer saline (pH 7.2). The morphology of oocytes was studied and was classed into grade I, II and III based on morphological appearance and presence of cumulous oophorus cells. The diameter of oocytes was measured with the help of occular micrometer. The average number of oocytes recovered from right ovary was slightly more than left ovary and the percentage of grade I oocytes recovered from right ovary also were more than left ovary. The diameter of oocytes with zona pellucida did not differ significantly in between I and II grades while it differed significantly (P0.01) when these two grades of oocytes were compared with the grade III. The diameter of good quality oocytes is significantly (P0.01) larger than poor quality oocytes. There was no significant difference in vitellus diameter in between the Grades I and IIoocytes but in grade III the same diameter differed significantly (P0.01) from grade I and II oocytes. No difference between zona pellucida thickness of grade I, II and III oocytes became evident under this experiment.
