Optimization of growth conditions of Arachniotus species on rice polishings for its protein enrichment

Protein enrichment of rice polishings was carried out through fermentation with Arachniotus species. Growth conditions of the microorganism viz., substrate treatment, supplementation of molasses to the growth medium, determination of carbon to nitrogen ratio, determination of time for supplementation of molasses, supplementation of corn steep liquor to the growth medium, determination of time for supplementation of corn steep liquor and harvesting time were optimized. Defatting of rice polishings had non significant effect on protein yield by the organism. Acid treatment of substrate could not improve the protein yield. Supplementation of molasses from 1 to 5% significantly depressed the protein yield by the organism at all the levels as compared to the control (without molasses). Carbon to nitrogen ratio 10:1 was found optimum for maximum yield of the biomass protein by the organism. For 1% molasses supplementation, 48 hours of fermentation was found optimum time. Supplementation of corn steep liquor at the rate of 0.5% was found good for maximum protein yield by the organism. Like molasses, 48 hours of fermentation was found optimum time for supplementation of 0.5% corn steep liquor. Best time for harvesting of biomass protein produced by the organism was found to be 72 hours. Through fermentation of rice polishings with Arachniotus species under the optimum conditions, the true protein of rice polishings was increased from 10.93 to 17.13%.
