Urinary excretion of acetylsalicylic acid in healthy human beings (male)

The concentration of acetylsalicylic acid as free saucy is acid was measured colorimetrically and absorbance was noted at 530 nm in urine of twelve (12) healthy male volunteers after an oral administration of 2x300 mg tablets. The urine samples were collected at 30, 60, 120, 180, 240 and 600 minutes after administration of drug. Aspirin is an acidic drug and readily absorbed from gastrointestinal tract and excreted in the basic urine. Excretion of free salicylic acid was extremely variable and depends upon pH of urine. The results show that the mean plus minus SE values for pH of urine excreted was 6.175 plus minus 0.18, for concentration of salicylic acid in urine was 37.55 plus minus 8.61 mug/mL and for diuresis was 0.028 plus minus 0.006 ml min-1 kg-1. The rate of excretion was 0.849 plus minus 0.226 mug min-1 kg-1, Amount of drug excreted as free salicylic acid was 5.40 plus minus 1.15 mg, percentage of dose excreted was 0.92 plus minus 0.19. The mean plus minus SE values for the cumulative amount of dose excreted in urine of male volunteers was 17.74 plus minus 2.91 mg cumulative percent of dose excreted was 5.47 plus minus 0.778 %. There is significant positive correlation between rate of excretion and pH of urine. Many factors can influence individual's response to drug including age, sex, disease of organs (kidney, liver), food and chemical conditions so the present study comprehensive evaluation of drug under obtain pharmacokinetic parameters on regimes of drug could be based.
