Study on preservation of croton (Codiaeum variegatum) callus in liquid nitrogen

Cryopreservation of croton callus in liquid nitrogen with different methods were as follows: stored in liquid nitrogen suddenly, using of cryoprotectant with 5 percent glycerol 5 percent glycerol storage in liquid nitrogen suddenly, using of cryoprotectant with 5 percent glycerol and decreased temperature before storage in liquid nitrogen, using of cryoprotectant with 5 percent glycerol and decreased temperature storage than washed with liquid medium after storage, using of cryoprotectant with 5 percent glycerol and decreased temperature before storage then increased temperature after storage, and using of cryoprotectant with 5 percent glycerol and decreased temperature after storage then increased temperature and washed with liquid medium after storage. The results showed that the best cryopreservation method of croton callus was the use of 5 percent glycerol and decreased temperature before storage then washed with liquid medium after storage in liquid nitrogen, which resulted in a slightly growth of callus after 50 days.
