Experiment was conducted to study the in vitro culture of pointed gourd during the period from April to August, 1996. Cotyledon rescued from physiologically mature seed (PMS) and immature seed (IMS) of pointed gourd were used as explants. Cotyledon excised from PMS responded very well in all culture conditions. Plants regenerated from these cotyledon ranged from 62 to 92 percent in different combinations. Plants regenerated from these cotyledon ranged from 62 to 92 percent in different combinations of media. It was highest in MS+1 mg/l BA and lowest in hormone free MS medium. The establishment rate of regenerated plantlets varied from 61 to 94 percent in vermiculite and 70 to 90 percent in soil media. Plantlets obtained from MS+1 mg/l NAA showed better performance in both media.
