Simple purification method for microcystins by low-pressure liquid chromatography

Cyanobacterial hepatotoxin microcystin (MC) causes problems in eutrophic rivers, ponds, and lakes throughout the world. More than 60 variants of microcystins have been reported, and the number will increase in future. Developments of large- scale purification method are required for the study on their chemical characters, structures and for the application of them to biochemical reagents. The large-scale purification method of four major microcystins from laboratory strain Microcystis SMST3, was studied with DEAE low-pressure liquid chromatography. The addition of EtOH to the eluents (20% EtOH) on DEAE chromatography gave high resolution, resulting in 96 and 88% purity with 65 to 70% recovery for MC-LR and -LW, respectively. Although MC-LY and -LF were not separated by DEAE chromatography, it was remarkable that DEAE chromatography was superior to reversed-phase chromatography for the separation of MC-LW and -LF. It is concluded that a combination of the present method and other chromatography becomes one of the practical approach for the purification of various MCs.
