In vitro selection and rapid screening of high-lysine mutants in rice

Lysine is the first limiting essential amino acid for optimal nutritional quality in rice grain. Somaclonal variation was the source of mutation in our breeding program. Biochemical selection was conducted using 1 mM S-(2-aminoethyl) cysteine followed by 5mM lysine plus threonine in the callus subculture medium. The lysine content in the endosperm of all progenies regenerated from the surviving calli was higher than that of their donor cultivars. These elevated levels of mutants were successfully transmitted to the M4 generation. Absorbance at 570 nm in the ninhydrin color test was significantly correlated with lysine content and the composition of rice endosperm.
