Identifying subspecies-specific DNA markers in rice
2003
Chin, J.H. | Koh, H.J.
Subspecies-specific random amplified polymorphic DNA (RAPD) markers using 30 varieties (15 japonica, 15 indica) of various origin were identified. Of 266 random primers tested, 25 primers produced 31 subspecies-specific bands: 11 bands were present only in japonica, 19 bands only in indica, and 1 band only Tongi-type varieties. Rice varieties were classified into two discrete subspecies groups by subspecies-specific markers. The SPV (subspecies-prototype variety) concept was introduced. Varieties could be classified by SPV index based on the genetic similarity to SPV using subspecies-specific markers. Breeding lines from crosses between Dasanbyeo (indica) and TR22183 (japonica), and Dasanbyeo (indica) and Seosan2 (japonica), were analyzed by selected subspecies-specific markers. All the lines turned out to be genetically closer to the indica parent, Dasanbyeo.
