Genetic diversity in seed storage proteins of Bangladeshi rice cultivars

A total of 467 Bangladeshi rice cultivars were analyzed for endosperm storage protein glutelin and prolamin compared with japonica and indica varieties Kinmaze and IR36. Screening was done by observing the profiles and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) on single-seed basis. A wide variation, in both banding pattern and staining intensity of polypeptide bands, was observed for glutelin and prolamin seed storage proteins. In the glutelin acidic subunit, seven different types were found based on apparent molecular size and staining intensity of three major polypeptide bands (alpha-1, alpha-2 and alpha-3), whereas two kinds of variation were identified at the alpha-4 level depending on molecular size. A great variation was also detected in the case of prolamin and the varieties were divided into nine arbitrary groups.
