Variation in seed storage proteins of Pakistani rice germplasm

Rice, the second most important crop in Pakistan, was investigated for endosperm storage proteins. Four hundred seventy-five accessions, obtained from the Ministry of Agriculture, Fisheries, and Forestry (MAFF) genebank of Japan, underwent SDS-PAGE and IEF analyses. A wide variation was found in local rice cultivars of Pakistan for the two major storage proteins, glutelin and prolamin. Prolamins showed four types of variation by SDS-PAGE, whereas glutelin revealed six types of variation. For proglutelin (57-kDa polypeptide), two types of variation were recorded in comparison with IR36 and Taichung 65. Some 90.5% of high 57-kDa polypeptide (57H) spontaneous mutants were distributed in low-altitude areas of the Punjab region. The IEF analysis of extracted glutelin also showed variation based on the intensity and presense or absence of the respective bands. The 57H mutation (controlled by a single recessive gene, i.e., glup3 gene) was found to be located between RFLP markers C1238 and R2370 within a 5.3-cM distance on chromosome 4. The observed variation for low prolamin and the glutelin variation may be useful in improving the nutritional value of rice.
