In vitro culture through nucellus tissue of Citrus reticulata cv. Shogon

Nucellus tissue of Citrus reticulata cv. Shogun were cultured on Murashige and Tucker medium (MT.) which supplemented with 0-2 mg/l. Kinetin (Kn.) and 0-1 mg/l. indole-3-acetic acid (IAA.). The result showed that nucellus tissues were developed to be small green tissue after 21 days. The development of nucellar embryo and plantlet were decreased when increased Kn. concentrations. IAA. at 0.5 mg/l. gave the highest differentiation percentage at 45 percent after 90 days. On every modified medium, nucellus tissue can differentiation into nucellar embryos and plantlets except at 2 mg/l. Kn. which can differentiated only plantlet. For multiple shoot induction, nucellus embryos were cultured on MT. which supplemented with 0-10 mg/l. Kn. It was found that Kn. at 10 mg/l. gave the shoot number of 7.14 shoots per piece with average shoot length of 0.72 cm. within 30 days. For root induction, shoots from MT. at 8 mg/l. Kn. were cultured on MT medium with reducing ammonium nitrate (NH4NO3) and potassium nitrate (KNO3) to 0 and 100 percent and in both medium with 0-1 mg/l. IBA. It was found that shoots from MT medium with 1 mg/l. IBA. gave highest root percentage at 40 percent while shoots cultured on both modified medium had lower root percentage with yellowish shoots. The survival percentage of plantlets were 100 percent after transplanting 50 days in nursery.
