Tissue culture of watermelon, Citrullus vulgaris

Various explants excised from seedlings of Citrullus vulgaris were cultured on Murashige and Skoog (MS) medium containing 15 percent coconut water and various concentrations of BA (6-benzylamino purine). Hypocotyl explant provided callus but no shoot formation on the medium with BA at concentration 0.5-4.0 mg/l. Cotyledon and shoot tip explants could be induced to form multiple shoots by culturing on MS medium containing 15 percent coconut water and BA at concentration 0.5-4.0 mg/l. Maximum number of shoots from cotyledon and shoot tip explants were obtained by inducing with BA at concentrations 1.0 mg/l for cotyledon and 2.0 mg/l for shoot tip. The average shoots obtained from cotyledon and shoot tip were 7.5 and 7.0 shoots per explant respectively. Rooting was successful in excised shoots grown on both MS medium containing NAA (naphthalene acetic acid) 0.5-1.0 mg/l and growth regulator-free.
