Variability of nitrogen efficacy in alfalfa, Medicago sativa L. clones

Efficacy of nitrogen fixation in alfalfa, Medicago sativa L., clones was tested in aim to determine nitrogen fixation variability depending on bacteria Sinorhizobium meliloti genotypes. Regenerated plants were inoculated by two different active strains of Sinorhizobium meliloti (218 and 224). The intensity of nitrogen fixation was determined in one clone genotype of each of the following alfalfa cultivars; K-22, K-28 and Mediana. Standard microbiological methods, plant growth in controlled environment and micropropagation from apical and lateral buds in vitro culture were employed. The inoculation clones were found to utilize N2 efficacely with different degree. Determination plant variability inside clone were 10-16% of coefficient variation (CV) for dry matter and total nitrogen, but for fixed nitrogen were some higher. A bi-factorial analysis of variance revealed a significant effect of the bacterial strain on nitrogen fixation variability. This effect was considerably higher than the effect of plant genotype or the interaction between symbionts of different genotypes, which proved another important influence.
