Development of vitrified cat oocytes and effects of glucose supplementation on in vitro development of embryos

In the first experiment, the effects of glucose supplementation on in vitro development of cat zygotes were investigated. The zygotes derived from in vitro maturation and fertilization were cultured for 72 h in modified Earle's balanced salt solution supplemented with or without 1.5 mM of glucose and then cultured for further 5 days. The supplementation of glucose significantly decreased the percentages of morula and blastocyst formation. In the second experiment, the development of cat oocytes cryopreserved by vitrification method was investigated. In vitro matured oocytes were vitrified and warmed by the straw and drop methods, and then fertilized in vitro. The proportion of survival of oocytes vitrified-warmed by the drop method was significantly higher than that of oocytes by the straw method. However, there were no significant differences between the two groups with respect to the cleavage and development to the morula stage. These results indicate that the glucose supplementation during incubation of embryos at the early stage has a negative effect on the embryo development. Since the development of vitrified-warmed oocytes was very low, further studies are required to improve the survival and developmental competence of cryopreserved cat oocytes.
