Nuclear localization of NF-KB in mouse embryos at the 1-and 2-cell stages, and effects of N-acetyl-L-cysteine and pyrrolidine dithiocarbamate on their development in vitro

Nuclear Factor kappa B (NF-kappa B) is a transcriptional factor that controls the expression of a number of genes related to immune responses, embryonic development and cell death and so on. In mouse preimplantation development, it has been reported that RelA, one of the subunits of NF-kappa B, is expressed in mouse eggs and embryos from the metaphase II (M II) oocyte to the blastocyst stage and that nuclear translocation of RelA, evidence for NF-kappa B activation, was observed at the 1-cell stage. In this study, immunofluorescence analysis showed that nuclear translocation of RelA, was observed not only at the 1-cell stage but also at the 2-cell stage. Moreover, we observed RelA punctate localization in nucleoplasm of embryos from the 1-cell to 2-cell stages and the nuclear dots were enriched conspicuously in the 1-cell and the late 2-cell embryos. These results suggest that NF- kappa B is activated in embryos from the 1-cell to the 2-cell stages, and nuclear dots in their nucleoplasm may be "enhanceosome", since it has been reported that transcription factors including NF-B and coactivators form punctate structures called "enhanceosome" in association with particular promoters in the nucleus.
