Chromosome doubling in Dendrobium anosmum using colchicine

Genetic traits of tetraploid homozygous plants are desirable in breeding. Occurrence of spontaneous chromosome duplication has been reported. It is the object of this study to determine the efficacy of autoclaved colchicines solution in chromosome doubling in homozygous lines of orchids. Dendrobium anosmum _giganteum_ protocorms were treated with 0.1% colchicines solution for 8, 9,11,13,15 and 17 days on a notary thumbler (at 1 rpm). Phenotypic aberrations were observed on all the treatments. This included squat plants, thicker leaves and stems, albinism and visual chimeral production (variegation and thickening of some sectors). Isolation of individual mutants was done by growing the Callus points. To screen the voluminous samples, Guard cell size will be used to predetermine chromosome doubling before the actual chromosome counting.
