[Creation of tetraploid forms of red clover (Tr. pratense L.) by the method of clonal micropropagation in vitro]

The method of creation of red clover (Trifolium pratense L.) tetraploid forms in the process of clonal micropropagation in vitro culture using the direct regeneration effect after colchicine treatment was described. Germs epicotyls and axillary buds of mature plants were used as the explants. Colchicine for the axillary buds was added into agarized nutritive substratum B5 that made it possible to create the tetraploids using the initial plants as the control ones. Quantity of the developed tetraploids depended on the genotypes of initial plant and on the amount of explants taken from the plant. Sterile epicotyls colchicinized by vacuum-infiltration were used for the increasing of number of diploid population transferring to the tetraploid level. The amount of pure tetraploids from the primary diploid population was almost 95%. Total number of tetraploid population was higher than the diploid one due to the high coefficient of clonal micropropagation. Creation of polyploid forms of red clover by means of clonal micropropagation made it possible to shorten the terms of polyploid population creation up to 6-7 months, increase the quantity of initial diploid population and realize the assessment and selection of initial tetraploid populations in winter period. The observed method made it possible to create tetraploid analogues of diploid genotypes on the level of solitary plants on the same genetic basis
