A study on tolerance and adaptability of sugar beet tissues of different ploidy levels to salt stress conditions in the regeneration medium

Adaptation of sugar beet tissues at different ploidy levels to salt stress condition of increasing sodium chloride levels in the regeneration medium was studied using sterile PGoB medium containing 0.9 % agar and 3% sucrose, BA and NAA at 0.5 and 0.2 mg/l respectively. Experiments were conducted with four levels of salinity: of no-salt, 100, 150 and 250 mM. For each 15 explants were inoculated in each Petri plates at three replications and were kept at 25؛ ± 2؛ C and 2000 Lx for a period of four weeks. Number of regenerated shoots was recorded. A factorial design with two variables was used and DMRT was performed. Results of ANOVA showed that in spite of general tolerance of tissues to salinity, differences observed for lines, stress levels and the interaction of these factors were significant. Line 7233 was more tolerant and 261 and 9597 had identically less tolerant. An experiment was conducted to study the adaptability of sugar beet zygotic embryos to stress conditions. Sterile whole seed and naked embryos from lines: 9597(salt sensitive) and 7233(salt tolerant) were cultured in water agar with the same salt levels. Naked embryos of both lines sprouted in both stress levels where as their whole seeds did not. There were significant differences between the plant material, salt levels and their effects upon each other. Furthermore seed advancing experiment with eight hours Thiram (0.2%) treatment and gradual decrease of seed humidity at 25؛ C was performed. Treated seeds of all three lines germinated in stress conditions. Results showed that lines, stress levels and their effects upon each other were significantly different. Line 7233 was most tolerant followed by 9597 and 261. The benefit of seed 'Advancing ' treatment for tolerance to salt stress conditions was demonstrated.
