In vitro fertilization of gilt oocytes by subzonal sperm injection

Ovulated oocytes were recovered from hormonally stimulated prepubertal gilts 40-43 hours after hCG application by flushing oviducts surgically or after slaughter. Only intact oocytes with extruded first polar bodies were selected for subzonal sperm injection (SUZI). Five to ten spermatozoa prepared by swim-up method were injected into the subzonal space of selected oocytes. Successfully injected oocytes were cultured in vitro for 18-20 hours, fixed, stained and evaluated for the presence of second polar bodies and two pronuclei. From 374 oocytes selected for SUZI, 352 survived injection and 44 (12.5%) oocytes were fertilized.
