Effects of FF-MAS on maternal and paternal pronucleus development in porcine embryos produced in vitro

Follicles with a diameter of 2 to 5 mm were punctured with a needle and oocytes were transferred into BSA free NCSU 37 supplemented with 6 mM L-cysteine, 10ng/ml epidermal growth factor and 50 uM ss-mercaptoethanol with or without 10 % porcine follicular fluid. For the first 24 hrs 1 mM db-cAMP and 10 IE PMSG/hCG were added, together with either 1.0, 3.0, 10.0, 30.0 or 100 uM follicular-fluid-meiosis-activating sterol (FF-MAS). To see the reversibility of high dosage of FF-MAS on oocyte degeneration, nuclear maturation and sperm penetration, in some experiments the oocytes were cultured for the first 24 h in follicular fluid free culture medium enriched with 100 uM FF-MAS. This regimen was followed by 24 hrs culture of the oocytes in FF-MAS free medium with or without follicular fluid. The results were compared with those obtained after 48 hrs culture of the oocytes in NCSU 37 serum free medium supplemented with 100 uM FF-MAS. At 16-18 h post fertilization ova were fixed in acetic acid alcohol and 24 h later stained with lacmoid. We found, that inhibition of nuclear maturation and fertilization was reversible only in FF+medium. Presence of follicular fluid in culture media increased the number of advanced paternal and maternal pronucleus stages (PN4-PN5), which are typical for this time point of culture. High concentration of FF-MAS significantly reduced the synchrony of pronuclei in both, FF+ and FF- media.
