[Development of the method of molecular diagnostic of Erwinia amylovora on the basis PCR-RFLP analysis]

The proposed method of identification of Erwinia amylovora of the basis of polymerase chain reaction (PCR) with the further restriction fragment length polymorphism (RFLP) analysis made it possible to clearly identify the collective strains of Erwinia amylovora from the other bacteria including closely related species which could be found in inhabitation of studied pathogen. Strains E. amylovova 8507 and 9057, Ralstonia solanacearum 9049, Erwinia carotovora SCRI1043, Pseudomonas chlororaphus 494 as well as Erwinia carotovora 3-2, Rewinia chrysanthemi 3937 and Panthoea agglomerans were used in course of realized investigation. Primers for the amplification of hrpN E. amylovova gene with access to the applied sequences were presented. Amplification of hrpN gene fragments from studied strains was analyzed. Prospective sizes of the fragments in the conditions of restriction with various endonucleases of hrpN gene from Erwinia amylovora strain 1/79 amplificated by hrpN1-hrpN2 genes were presented
