Evaluation of the Efficiency of Indigenous Rhizobium leguminosarum bv. Viciae in Symbiosis with Lentil in Soils of Ardabil Province.

For evaluation of nitrogen fixation potential by indigenous isolates of lens nodulating rhizobia an experiment was carried out during 2003-2005. In the first year, 212 soil and lens root nodule samples were collected from different sites of Ardabil during three sampling stages. Soil samples were inoculated to lens sterile plants following a four step dilution procedure. Nodules were formed on the roots as well as the nodules were collected from the fields, were crashed and streaked on YMA plates followed by different subculturings to obtaining purified cultures. Seventy-six isolates were obtained in this step. The isolates were authenticated by a plant infection test. Forty-five isolates were able to form nodules on the roots of lens plants. Nitrogen fixation effectiveness of the isolates were evaluated in a RBC design including 45 isolates, N application (35 mg N/kg) and control (without inoculation and N fertilizer) in three replications. Most of the isolates (more than 60%) had good effectiveness for N2 fixation. Some isolates increased shoot dry weight by 5-12% and shoot N-content by 28-36% compared to N-fertilizered and control treatments, respectively. Twelve isolates were selected as superior and their efficiency were evaluated in a greenhouse experiment using a soil was dominant in province (Inceptisol) in a RBC design in three replications. Plant growth parameters including shoot dry / wet weight and N-content and seed yield were measured compared to a N-fertilized treatment (150 kg urea/ha). The results showed that indigenous isolates were able to increase the seed yield by 20-30% over control. Finally four isolates with highest effect on growth parameters were selected as superior isolates.
