Cytological and immunocytochemical studies on the effects of the fungicide tebuconazole on the interaction of wheat with stripe rust [Triticum aestivum L.]

إنجليزي. Effects of the fungicide tebuconazole, applied as a leaf spray after inoculation of wheat leaves with stripe rust, were examined by means of electron microscopy and immunogold labelling. Tebuconazole strongly inhibited the development of Puccinia striiformis in host leaf tissues and also markedly altered the structures of the hyphae and haustoria. These changes included increased vacuolation, irregular thickening of cell walls, necrosis or degeneration of cytoplasm and collapse of the hyphae and haustoria. Large differences were detected in P. striiformis-infected wheat leaves treated with fungicide compared with the untreated control. A very thick layer of wall appositions and large encasements were usually formed in the infected fungicide-treated leaves, whereas such structures were never observed in the infected wheat leaves without fungicide treatment. In addition, chitinase, beta-1,3-glucanase and lignin were detected in wheat leaf tissues by immunogold labelling. Accumulation of the two enzymes and lignin increased slightly in infected leaves compared with uninoculated healthy leaves, while a much higher accumulation of the two enzymes and lignin was detected in infected wheat leaves after fungicide treatment. Our results suggest that tebuconazole not only inhibited development of P. striiformis in wheat leaves, but also enhanced structural and biochemical host defense reactions in the infected host

[Sono stati esaminati gli effetti del fungicida tebuconazolo, somministrato tramite irrorazione dopo l'inoculazione di foglie di frumento con ruggine striata, mediante microscopia elettronica e immuno-oro marcatura. Il tebuconazolo inibiva fortemente lo sviluppo di Puccinia striiformis nei tessuti fogliari dell'ospite e alterava pure marcatamente la struttura delle ife e degli austori. Questi cambiamenti consistevano in vacuolizzazione, ispessimento irregolare delle pareti cellulari, necrosi o degenerazione del citoplasma e collassamento delle ife e degli austori. Sono state notate differenze notevoli nelle foglie di frumento infettate da P. striiformis trattate con il fungicida rispetto al controllo non trattato. Nelle foglie infette trattate con il fungicida si formava di norma uno strato molto spesso di apposizioni e ampi rivestimenti, mentre tali strutture non sono state osservate nelle foglie infette non trattate con il fungicida. Inoltre, mediante immuno-oro marcatura, nei tessuti fogliari del frumento sono state identificate chitinasi, beta-1,3-glucanasi e lignina. L'accumulo dei due enzimi e di lignina aumentava leggermente nelle foglie infettate rispetto a quelle sane non inoculate, mentre è stato riscontrato un accumulo molto più elevato dei due enzimi e di lignina nelle foglie infettate dopo il trattamento con il fungicida. I nostri risultati fanno ritenere che il tebuconazolo non solo inibisse lo sviluppo di P. striiformis nelle foglie di frumento, ma potenziasse le reazioni di difesa dell'ospite in caso in infezione.]
