Identification of rice [Oryza sativa] varieties using DNA marker

In the present study, we tried to identify the rice varieties bred in Aichi Agricultural Research Center from other major commercial varieties in Japan by DNA analysis based on PCR method. 27 varieties of nonglutinous rice, 5 varieties of brewer's rice and 9 of glutinous rice were tested. PCR primers were designed by Ootsubo et al. (National Food Research Institute), Hayashi et al. (National Institute of Agrobiological Sciences) and Mizukami et al. (Aichi Agricultural Research Center). These primers were used for PCR analysis to detect the unique polymorphism among the tested varieties. Using these primers, all of tested varieties were identified from each other. Then, as the attempt to identify rice varieties more easily, we applied the multiplex PCR. By multiplex PCR using the primer mixture, which was selected from the results of former PCR analysis, the identification of rice varieties was also succeeded. The multiplex PCR method developed in this study made it possible to identify the variety even if the materials were only one rice grain. From these results, PCR and multiplex PCR method was shown as the useful tools to identify the rice varieties bred in Aichi Agricultural Research Center.
