Development and evaluation of DNA markers for Japanese malting barley [Hordeum vulgare] breeding

DNA markers were developed for selecting lipoxygenase-deficient lines and published DNA markers were evaluated for four breeding characters to develop an efficient system for malting barley breeding. For the efficient selection of lipoxygenase-deficient lines which are effective to keep beer flavor and form stability, cleaved amplified polymorphic sequence (CAPS) and genotype-specific PCR markers were developed. DNA markers for serine protease activity (Cxp1), barley yellow mosaic virus resistance (rym5), powdery mildew resistance (mlo) and winter habit (Vrn-H2) were evaluated. Based on the analysis of the genotype variation among Japanese cultivars, we found that the usability of the markers for rym5, mlo, and Vrn-H2 has limitation in Japanese malting barley breeding.
