Aim of this study is optimalization methods of isolation Cucumis spp. pollen grains. These isolated pollen grains can be utilized for in vitro pollination within the genus Cucumis. This type of pollination is useful for interspecific hybridization to transfer some specific genes for various resistances from wild Cucumis spp. genotype to cucumber genotype. Successful interspecific hybridization will be used in cucumber breeding. The main influence to pollen grains has type of isolation, type of isolation (washing) solution, type of cultivation media and others. For these experiments were used two accessions of Cucumis spp. - C. sativus and C. melo. Three types of isolations, three types of isolation (washing) solutions and two types of cultivation media for pollen grains of these two accessions were tested. There were observed percentages of pollen grains viability, of occurred pollen tubes and arisen embryos (embryo-rescue cultures). Various methods of isolation were compared and their utilization for successful in vitro pollination was evaluated.
