Effect of medium change to percentage of viable BHK21C13 suspension cells stored at 4 deg C

Large scale cultivation of BHK sub(21) C sub(13) cells for FMDV production takes about 20 days starting from growing seed cells in a 500-ml culture glass bottle to 2 L and 5L Woulff bottle, then 40 L, 90 L, 300 L, 2,000 L and 4,000 L fermentors. Thus storage of BHK sub(21) C sub(13) suspension cells in a 300-L fermentor at 4 deg C about 2.5*10**(6) cells/ml would reduce the re-cultivation time in case bacterial contamination or poor cell growth problem occurs. In this study change of the old medium helped sustain the viability of the stored suspension cells. To maintain enough viable cell number required for cultivation in 2,000 L and 4,000 L fermentors, the suspension cells can be kept up to 40 and 20 days, respectively.
