Comparison of membrane filtration culture to Karmali selective media for the isolation of thermophilic Campylobacters in broiler intestine at slaughter

Total of 180 randomly-sampled broiler intestinal specimens were collected on three separate occasions, from 2 slaughterhouses. Eighteen from 180 samples were sampled and studied for normal colonization of thermophilic Campylobacters. It was found that 17/18 were positive for thermophilic Campylobacter organisms, which the average counts were at 3.1*10**(8), 4.3*10**(7) and 4.3*10**(6) colony forming unit/gram (CFU/g) (53 percent, 35 percent and 12 percent respectively). The concentration of thermophilic Campylobacters that could be detected by membrane filtration method were at the average of 3.1*10**(3) (n = 8 samples) while 3 samples each were at the average of 6.3*10**(1), 3.2*10**(2) and 5.8*10**(4) CFU/g. A comparison was performed of selective enrichment Preston broth culture in combination with 0.45 micro m membrane filtration culture onto blood no. 2 agar versus Karmali selective culture as recommended by the standard method (ISO/DIS 10272, 1994). Of the 180 samples tested, 74 were positive by both modified selective enrichment membrane filtration and Karmali selective culture, 29 were positive by membrane filtration alone 14 were positive by Karmali selective culture alone, and the remaining 63 samples were negative by both methods. Using membrane filtration culture the number of positive for thermophilic Campylobacters was greater than those of the Karmali selective media (103 and 88 samples respectively). The membrane filtration culture was found to be useful in elimination of the commensal flora in the broiler intestinal samples. Furthermore the colonies on blood no. 2 agar without antimicrobial agents were much larger than those from Karmali selective media that may be beneficially for further studies.
