[Influence of plasmid abundance with genes of carotenoid biosynthesis on production of beta-carotene by Escherichia coli strains]

Research of interaction between the level of beta-carotene production by Escherichia coli strains and abundance of plasmid with genes of carotenoid biosynthesis (crt-cluster) of Pantoea agglomerans 206 bacterium was realized in the conditions of the Republic of Belarus. It was demonstrated that in case of ten-fold increasing of plasmid copy number the increasing of strain productivity took place only in 1,7-2,2 cases for Escherichia coli DH5alpha and E. coli XL1-blue, respectively. Apparently, the weak dependence between the beta-carotene and plasmid abundance could be explained by the presence of some limiting factors (limited number of isoprenoid precursors, impossibility of accumulation of large number of carotenoids in cell membranes) in E. coli bacterium. Research results revealed that the production of carotenoid pigments by E. coli DH5alpha strains in 1,5 times exceeded their production by E. coli XL1-blue ones. That obtained results proved that E. coli DH5alpha could be more preferable host for the plasmid with the genes of beta-carotene biosynthesis in comparison with E. coli XL1-blue
