Detection of Infectious Bronchitis Virus by a rapid hemagglutination test using Neuraminidase enzyme

Rapid hemagglutination (HA) test was used for detection of infectious bronchitis virus (IBV) in infected allantoic fluid (AF) of SPF embryonated eggs. The virus was detected by observation of HA activity after neuraminidase treatment. Thirty IBV suspected cases were examined by rapid HA test. Suspected materials were inoculated in SPF embryonated eggs via chorioallantoic cavity. Harvested AFs were treated with neuraminidase enzyme. The IB virus was detected by induced hemagglutination of chicken red blood cells. The specificity of rapid HA test was examined with other non-hemagglutinating avian viruses including infectious bursal disease virus (IBDV), infectious laryngotracheitis virus (ILTV), fowl pox virus (FPV) and reovirus. The sensitivity of the test was compared with reverse transcriptase-polymerase chain reaction (RT-PCR). The results showed that this test was specific and had a sensitivity of 71.4% in the first passage, 85.7% in the second passage and 92.8% in the third passage for IBV detection.
