Development of a DNA marker tightly linked to the root-knot nematode [Meloidogyne] resistance gene, Mi, in tomato [Lycopersicon esculentum]

We tried to develop a DNA marker linked to the root-knot nematode resistance gene, Mi, in tomato. 1000bp and 500bp fragments were amplified by PCR reaction with a set primers (Forward:5'-AGCACTTCAGGGGCTCGAAAC-3', Reverse: 5'-CACATCCTTGGTCGACCGTG-3'). We cleaved these amplified fragments by restriction enzyme MseI. We detected a 350bp fragment in nematode resistant homozygote lines and a 500bp fragment in sensitive lines. It also both fragments were detected in resistant heterozygote lines. We developed codominant marker.
