Evaluation of the punched aluminum metal sheet as the instrument for the cryopreservation of porcine embryos

The objective of this study was to determine the efficiency of cryopreservation using the instrument of our own making by aluminum punching metal sheet (PM) for porcine embryos. Embryos at expanding blastocyst stage were vitrified by using open pulled straw (OPS) and PM methods. Viability of vitrified/warmed blastocyst was estimated in vitro, by the percentage of embryos that developed after 24 h and hatched by 48 h. The quality of survival embryos was evaluated by the total cell number. The survival rates after 24 h culture in OPS and PM were 83.0% and 87.1%, respectively. The hatching rate by 48 h culture in OPS and PM were 76.8%and 78.3%, respectively. The total cell number of survival embryos in OPS and PM were 99.8+-9.5 and 97.9+-22.1. These results indicate that there is possibility as practical method for vitrification using PM.
