Antibacterial and antioxidant activities of sericin powder from eri silkworm cocoons correlating to degumming processes

This work was aimed to demonstrate the importance of degumming process in producing eri sericin powder with relatively high antibacterial and antioxidant activities. In a preliminary study, 90-min boiling in 0.25% Na2CO3 was selected as a promising method according to its high extraction efficacy and moderate degradation effect on sericin molecules. The molecular heterogeneity of eri sericin powder obtained from aqueous solution containing Na2CO3-degraded sericin polypeptides, as well as its antibacterial and antioxidant properties were compared with those of boiling water-degraded eri sericin powder and sericultural wastewater-produced eri sericin powder. The results conclusively indicated the superiority of the Na2CO3-degummed eri sericin powder in terms of its biological activities. These evidences were found in correlation with its unique characteristic of molecular size distribution in such a way that it lacks hydrolysate of polypeptides larger than 66 kDa. Other eri sericin powders in the present study gave relatively lower antibacterial activity, with exceptions for water degummed eri sericin against Gram-positive bacteria, and antioxidant activities while possessing abundant polypeptides with sizes up to 97 kDa.
