Syntheses of diester phthalates with potential antidiarrheal activities

A potential anti diarrheal agent,bistridecyl phthalate 1, isolated from Sapal (Drimys piperita Hook f.) leaves, was synthesized via Schotten-Baumann reaction. Using the charcoal tracing method, an antidiarhheal bioassay,it was found that compound 1 had comparable activity with the positive standard Imodium while phthalic acid,its hydrolized form, had no activity. The absence of activity of phthalic acid led to the conclusion that the long alkyl chain of the compound contributes to the observed antidiarrheal activity. With these results,the syntheses of analogs of compound 1 with varying alkyl chain lengths, compounds 2 to 8, were also conducted. Results of the bioassay showed that there is an increasing percent inhibition,as the alkyl chain gets longer until 13 carbons and then the activity dropped, as the length of the alkyl chain was further increased. Based on these results,it was established that the original compound 1 had the highest antidiarrheal activity and thus, for this particular series of compounds, the 13-carbon chain was the optimum length.
