Marker-aided selection in the development of transgenic fruit and shoot borer resistant Philippine eggplant

Eggplant (Solanum melongena L.) is an important vegetable crop in Asia. Eggplant fruit and shoot borer (EFSB), Leucinodes orbonales Guen is one of the destructive pests of eggplant. Fruit and Shoot Borer Resistant (FSBR) eggplant Philippine varieties are being developed through conventional backcrosses breeding. Two Philippine varieties, Dumaguete Long Purple (DLP) and Mara were initially crossed with the Indian line developed by Mahyco expressing the Cry1Ac gene against EFSB (Mahyco EE-1). Subsequent backcrosses were made through marker-aided selection. Bt positive individuals from BC2F1 and BC3F1 generations were selected based on qualitative ELISA strip test using the DesiGen Xpresstrips TM kit developed by Mahyco. Xpresstrips TM kit is a lateral flow device used by to quickly identify Bt Cry1ac-positive plants thereby genetic and generational stability of the Bt cry1Ac protein expression in both crosses. Chi-square data of the segregation in BC2F1 and BC3F1 generations conformed to the expected 1:1 Mendelian inheritance in a backcross population indicating that the cry1Ac gene behaves a single dominant gene and is stably integrated in the genome.
