Molecular characterization of introgression from Oryza glaberrima into rice (Oryza sativa L.) using molecular markers

Of the two cultivated rice species, O. sativa is high yielding and grown worldwide whereas O. glaberrima is low yielding and cultivation is only limited to western Africa. However, O. glaberrima is an important source of genes for weed competitive ability and tolerance to biotic and abiotic stress. A large number of advanced backcross progenies have been produced from crosses of O. sativa and O. glaberrima using O. sativa as recurrent parent at the International Rice Research Institute (IRRI). A set of 112 advanced introgression lines (ALs) derived from an O. sativa (IR69502-SRN-3-UBN-1-B) and O. glaberimma (TOG6216) cross were used to detect introgression of segments from O. glaberimma. Two hundred ninety two SSR markers that were distributed at a distance of approximately 10cM across the 12 chromosomes were used . Molecular analysis showed a 66.3% genome of the recurrent O. sativa parent, and 23.0% and 6.5% with O. glaberimma/O. sativa genome. Non-parental bands were also detected in the AILs. As many as ten markers did not detect introgression from O. glaberimma in chromosomes 3, 4, 6, 7, 8, 11 and 12. One of the markers (RM479), detected introgression in 86 AILs-Introgressed segments ranged from 0 to 2 in Chromosome 11 and 1-7 in Chromosome 6. The data generated are important in developing chromosomal segment substitution lines (CSSLs) and in the construction of molecular maps.
