Endogenous levels of IAA and ABA on developing bulbils of Agave ferox L.

Bulbils development in Agave ferox L. was studied for changes in endogenous level of conjugated IAA and free IAA and ABA. Bulbils were grouped as A to N according to their position on the stalk from top to the botton. From each group, bulbils were weighed, length was measured and crushed in liquid nitrogen in triplicates. To estimate endogenous level of IAA and ABA, antibodies were raised in rabbits till sufficient titer achieved. The serum was separated and antibodies were purified using ion-exchange chromatography. Conjugate IAA was hydrolyzed to free using 2M KOH. Indirect ELISA was performed to estimated endogenous level of IAA and ABA. It was observed that during early stage of development of bulbils, IAA level gradually increased till 6th group (F) and declined there after. In contrast, conjugate IAA levels were higher during later stages suggesting that free IAA gets converted to the conjugated form with the developmental stages. Free ABA level was low during initial stages and increased with age of the bulbils. The probable roles of IAA and ABA in bulbils development are discussed.
