Evaluation of the effect of late bight-resistant potato plants on the structure of bacterial associations in soil

The compositions of microbial associations isolated from soils where nontransgenic and transgenic lines of potato varieties Lugovskoi, Charodei, and Golubizna had been grown were under study. The transgenic lines are in possession of thaumatin gene (Tau2) improving plant resistance to main fungal and bacterial diseases. The analysis was based on denaturing gradient gel electrophoresis of total amplificates of 16S rRNA gene fragments and analysis of libraries of nitrogen fixation gene (nifH) fragments. Electrophoretic profiles consisted of 20 or more discrete stripes. There were nonsignificant differences in intensity of stripes, but not in their number. Diversity of diazotrophe sequence types of transgenic and nontransgenic varieties was equally. The minor differences detected in nitrogen-fixing microorganism diversity between control and transgenic lines of Lugovskoi and Golubizna varieties are associated probably with inadequate sample number. Neither method revealed significant differences in dominant components of the microbial associations isolated from soils with control or transgenic plants. It is concluded that transgenic plants have no considerable influence on qualitative composition of soil diazotrophic association.
