Xanthomonas axonopodis pv. glycines causes bacterial pustule of soybean, which infects severe losses of susceptible cultivars in Thailand and many parts of soybean production areas where the climate is warm and high humid. We have reported in the previous study that rpfF gene is regulated in pathogenicity but has not reported in X. axonopodis pv. glycines. Marker exchange mutations in the rpfF gene of X. axonopodis pv. glycines led to change qualitatively phenotypes. The mutant strain led to reduce disease severity. Beside, rpfF gene is regulated in increase bacterial population and epiphytic fitness on soybean leaves. The relation between X. axonopodis pv. glycines (rpfF mutant) strain T3A that rpfF mutant and X. axonopodis pv. glycines No12-2 (wild type) showed the mutant strain has efficacy to colonize on soybean leaves and to reduce the disease severity of X. axonopodis pv. glycines No12-2 (wild type). This study may provide new basis knowledge for the development of plant disease protection or apply for reduced virulence of the pathogens.
