صينى. 确定干扰素γ(IFN-γ)对妊娠早期大鼠下丘脑-垂体-卵巢轴中IGF-1表达的影响。将妊娠9 d的SD大鼠随机分为对照组、试验1组和试验2组,采用生殖道肌肉注射的方法,对照组大鼠生殖道肌肉注射生理盐水,试验组大鼠注射IFN-γ,其中1组剂量为2.5×104 U/只,2组剂量为7.5×10 4 U/只。各试验组均在注射48h后灌注取材、免疫组化SP法染色,光镜下观察。注射2.5×104 U/只和7.5×104 U/只的IFN-γ均能下调妊娠大鼠下丘脑室周核、视上核、视前大细胞核、视前内侧核、视前外侧核、视前交叉上核、弓状核,腺垂体,卵巢黄体及子宫蜕膜基质细胞和子宫腺中IGF-1的表达。IFN-γ对妊娠早期大鼠下丘脑-垂体-卵巢轴中IGF-1的表达具有下调作用,高剂量IFN-γ的下调作用更显著。

The study investigated the effect of interferon-γ(IFN-γ)on the expression and distribution of insulin-like growth factor-1(IGF-1)immuonore-active production in Hypothalamus-Pituitary ovary-Gonad axis of earlier pregnant rats in the post-implantation period. In this study the ultrasensitive immunohistochemistry SP method was used 9 d pregnant rats were divided into control group, experimental group one and experimental group two. The control group was injected physiological brine and the two experiment groups were injected 2.5×104 U and 7.5×104 U of IFN-γin vaginal muscular respectively. Experiment materials were collected after perfusion from each group which was injected 48 h ago, then stained by ultrasensitive immunohist -ochemistry SP method. And the results were observed and analyzed. The expressions of IGF-1 in pre-opticus medianus, nucleus supra-opticus, nucleus pre-opticus mango cellularis, nucleus pre-opticus medialis, mucleus pre-opticus lateralis, nucleus pre-opticus suprachiasmatic and arcuate nucleus of the hypothalamus, adenohypophysis, corpus luteum of ovary, stroma cells of uterus decidua and uterine gland were reduced after being injected 2.5×104 U and 7.5×104 U IFN-γ. The results suggested that IFN-γcould reduce the expression of IGF-1 immunological reaction production in Hypothalamus-Pituitary gland-Gonad axis. High dose of IFN-γshowed a much more significant down regulation.
