Here we present preliminary results on the assessment of the Y-chromosome diversity in Spanish goat, particularly focusing on the rare Bermeya goat breed. Three SNPs previously identified on the caprine AmelY and ZFY genes were genotyped using a RT-PCR protocol designed ad-hoc. Sample genotyped included 38 Bermeya males (20 from the Eastern and 18 from the Western subpopulations), 13 males from the Guadarrama breed, 2 from the Malagueña breed, 23 from the Murciano-Granadina, 6 from the Blanca Andaluza and 3 from the Payoya breed. Two different haplogroups were identified: C1, which was predominant in our sample, and C2 which was present in 3 Bermeya males belonging to both the Eastern and the Western subpopulations as well as in one Malagueña. The present analysis will be the basis of further characterisation of Y-chromosome diversity in Spanish goat using Y-specific microsatellite polymorphism.
