Isolation and identification of M.agalactiae and M.mycoides(LC) by culture and PCR from milk in Kordestan provience.

Contagious agalactiae(C.A.) is one of the most common disease affecting small ruminants.Mycoplasma agalactiae and mycoides larg colony are the agant of contagious agalactiae. C.A. is particularly widespread around the world. The disease in some animals have no signs so they are carriers and have difficulties in diagnosis of the disease.Culture of Mycoplasma need selective media and isolation of the bacteria need more time. Since infection spreads quickly,it is very important that specific and rapid diagnostic procedures are developed for its detection. The aims of this research was isolation of Mycoplasma agalactiae and mycoides large colonywith cultural and PCR methods in milk in Kordestan provience. In this research 367 milk sample collected. 20(5,5٪) out of 367 were positive in selective culture media.17(4,6٪) out of 367 were positive for PCR with universal primer of mycoplasma. 5(1,3٪) out of 367 were positive with mycoplasma agalactiae primer. Result of PCR with 367 milk samples with mycoplasma agalactiae showes that 11(3٪) of them were positive.Non of the samples were negative in culture and PCR for Mycoplasma mycoides large colony.PCR is the most promising due to its rapidity,its economical convenience and its sensitivity since it can detect few microorganism in clinical samples.It suggests that culture as an gold standard use besides PCR.
