Histopathology of Pneumocystis carinii in the lung of immunodeficient rats

Pneumocystis carinii or P.jiroveci is an opportunistic agent living in human and animal lungs. P.carinii was found to be an important patant pathogen causing pneumocytosis in immunosupressed hosts. In the present work, immune system of rats (Sprague dawley) was supressed by methyl prednisolone firstly. Then the infected rats were killed and impression smear was prepared from their lungs. The smears were stained by geimsa and Gomori';s methenamine silver (GMS) methods. Inaddition, the infected lungs were cut and tissue sections were prepared for histological study. The tissue sections were stained by H and E, geimsa and Gomori';s methenamine silver methods. Results indicated that, the infected lungs were found enlarged and heavy with a firm rubbery consistency. The principal finding on examination of lung section is the presence of the parasite cysts attached to the alveolar septa and hyaline foamy granular eosinophilic exudate in the honycomb from alveoli. The alveolar septa are thickened and there is typically interstitial inflammation with proliferation of plasma cells, congestion and intra-lobolary eodema in connective tissue. For detection of P.carinii organisms either in smear or in tissue section, the GMS staining was found more convenient.
