Study On Large Scale Production Of Autoclaved And Freeze - Thawing Inactivated Leishmania Major Vaccine Against Human Rural Cutaneous Leishmaniasis

Live promastigotes of wet rurel Leishmaniasis were grow then Harvested. They were separated from components of culture medium, without using merthiolate, through 5 times contrifugation and washing with saline solution. The resulted promastigotes (Promastigotes packed) were frozen at _lODe and maintained until use. three lots (lot10, lot 11 and lot 12) of live concentrated promastigotes were produced separately. lot 10 consisted of 1 sub-lots which were mixed and the volume increased to 400 ml by adding saline solution. 100 ml of the resulting promastigotes concentration was removed and divided into two equal parts which were inactivated through freeze thawing and autoclave method respective!y. They were delivered to the Ministry of Health, Education and Treatment. According to the reports, autoclaved L.major vaccine proved to be more efficient, therefore the remaining 300 ml of lot 10 was diluted with saline solution and the volume increased 4050 ml them the diluted promastigotes were autoclaved and delivered to the Ministry of Health, Education and Treatment. The autoclaved vaccine is enough for morethan 25000 volunteers. Now 8000 velunteers are vaccinated in endemic areas infected with L.major, L.tropica and L.donoveni in Iran (Esphahan,Bam) Pakistan and sudan, Packed promastigotes in lot 11 was showed and the volume increased to 300 ml and then Rept at -7 until use. lot 12 was maintaine intact at -7until use.
