Candidate genes in the major rice QTL for phosphate uptake Pup1

Phosphate uptake1 (Pup1) is a major quantitative trait locus (QTL) in rice that confers tolerance of phosphorus (P) deficiency in soil. The Pup1 region has recently been sequenced in the tolerant donor parent Kasalath. Within the Kasalath Pup1 region, 68 putative genes have been predicted by RiceGAAS. Detailed sequence analyses revealed that Pup1 is a hot-spot of transposon integration (750% transposon-related elements) with a complex genetic structure. In silico analyses of the predicted gene models failed to reveal any gene that is obviously related to P uptake or P metabolism suggesting that Pup1 confers tolerance via a novel mechanism or a regulatory gene. The authors are now characterizing and validating short listed candidate genes by RT-PCR expression analyses and other molecular approaches. In this context, the authors are analyzing lignification of roots of contrasting near isogenic Pup1 line (NILS) grown under combined drought and P stress. The data show large differences in root lignification in response to stress. However, differences between the contrasting NILS still need to be verified by quantifying the staining intensity of lignin.
