Construction of expression vectors of dap A gene to enhance Lysine content in rice endosperm

The improvement of the quality of protein in rice is a cost-effective way to combat the protein-calorie malnutrition in Asia. It has previously shown that expression of Corynebacterium glutamicum dihydrodipicolinate synthase (DHPS), and enzyme regulating the lysine biosynthetic pathway, increased the lysine content of canola and soybean seeds. In this paper, the authors described the construction of expression vectors of the C. glutamicum dapA gene under the control rice endosperm-specific promoters. The dapA gene was amplified from C. glutamicum and fused in frame with the chloroplast transit sequence (cts) of ribulose biphosphate carboxylase small subunit, (ss1) gene from maize. The resulting mcts-dapA gene was shuttled into the endosperm-specific gene expression vectors, yielding pC1300int-GluB1p-mcts-dapA and pC1300int-GluB4p-mcts-dapA. These construct are being transformed into rice cv. IR64 using Agrobacterium tumefaciens.
