Cellulose synthase as a major candidate gene in the large effect QTL for rice yield under drought stress

For rice yield under drought stress, a large-effect QTL 9DTY 12.10 derived from a cross between the varieties Vandana and Way Rarem was recently reported. The author used in silico interaction network analysis of the approximately 2-Mb QTL region on chromosome 12 to identify six candidate genes, one of which was cellulose synthase 10 (Ces A10). The Ces A gene complex is involved in primary cell wall synthesis. The spatio-temporal expression of each Ces A is not known however, some are involved in root hair elongation, which contributes to drought tolerance. To determine the role of Ces A10, molecular and phonotypic analyses was undertaken on the parental (Way Rarem, Vandana), control (Nipponbare) and four near-isogenic lines (NILs; +21, +83, -21, -83) derived from the cross. PCR-amplification and Southern hybridization indicated the presence of Ces A10 in Nipponbare, Way Rarem, and QTL + NILs but not in Vandana and QTL-, NILs. Cloning and sequencing of Ces A10 from varieties/NILs containing the gene indicated an identical sequence. Higher root biomass and root length was observed in QTL + NILs. Total root length measured using WinRhizo indicated that 75%of it comprised the fine roots. Further analysis also indicate4d better proliferation of the root hair in QTL + NILs after drought treatment. Similar results for Ces A10 function were obtained from 22 rice varieties screened for drought tolerance and then assessed for presence or absence of Ces A10. Difference in relative water content implicated earlier in driving the function of DTY 12.1, together with the present result suggests Ces A10 as a major candidate gene for DTY 12.1 function.
