Changes in the endocrine system might be a useful indicator of lead (Pb) exposure and its potential toxicity in animals. The study aimed at examining expression of proliferation- related (cyclin B1) and apoptosis-related (caspase-3) peptides in porcine ovarian granulosa cells after lead administration. Ovarian granulosa cells were incubated with/without lead acetate for 18 hours: group Max (0.5 mg Pb per ml), group A (1:1 dilution), group B (1:5 dilution), group C (1:7 dilution), group D (1:10 dilution) and the control group without Pb addition. Expression of cyclin B1 and caspase-3 were detected by immunocytochemistry and TUNEL assay. Lead was shown to be capable of accumulation in porcine ovarian granulosa cells. The expression of cyclin B1 and caspase-3 were influenced by experimental Pb administration. Percentage of ovarian granulosa cells containing cyclin B1 and caspase-3 was induced by Pb administration in all experimental groups. Our results contribute for knowledge of mechanisms of lead effect on porcine ovarian granulosa cells. These results show that lead can affects by pathway of proliferation and apoptosis of porcine ovarian granulose cells through intracellular substances as are cyclin B1 and caspase–3. These findings contribute to our knowledge of the effect of lead on ovarian function.
